Design Notes

1. Choose a linker when expressing a fusion protein of lasI and lacZ that ensures both of their functions. 2. The Stop Codon of lasI ORF should be mutated. 3. There should not be frame-shift mutations.


Source

It is cloned and modified from lasR-lasI signaling system in Pseudomonas aeruginosa. Sequences of lasI Promoter were obtained from the paper 'The quorum-sensing negative regulator RsaL of Pseudomonas aeruginosa binds to the lasI promoter' published in J Bacteriol (DOI: 10.1128/JB.188.2.815-819.2006). Sequences of related protein ORF were acquired from NCBI: https://www.ncbi.nlm.nih.gov/gene/881777.
